This will allow to propose the personalized therapy for patients with embryonic tumors and, thus, increase the effectiveness of complex treatment, improve survival rates and the quality of life. popup.description2 The WERI RB retinoblastoma sub-lines WERI-MRPS18-2, WERI-MRPS18-1, WERI-MRPS18-2-RB, as well as the A375 melanoma sub-line, expressing MRPS18-1-3 constitutively, were created and archived. The possibility of directed differentiation of the above-mentioned WERI RB retinoblastoma cell sub-lines in vitro was showed, using cocktails of chemicals, in osteo-, chondro- and adipo- lineages, that was confirmed by qualitative reactions to calcium ion deposits, negatively charged biopolymer molecules and triglycerides. Samples were prepared to determine the stability of mRNA of MRPS18 family genes in vitro in MCF7 cell cultures by qPCR, using actinomycin D, which is a fast-acting DNA intercatalyst and RNA polymerase inhibitor. Supramolecular complexes of antisense RNA against MRPS18-2 and control siRNAs on the surface of gold nanoparticles were prepared to study their effectiveness in cell cultures in which this protein is overexpressed, namely, A375 melanoma cells and WERI retinoblastoma cells. We assessed the expression patterns of the RB1 gene at the mRNA and protein levels in brain tumors. Heterogeneity in the expression pattern of RB1 was found in astrocytoma, glioblastoma and atypical meningioma samples, where 20 to 60% of tumors were positive, while in all embryonal tumors of the central nervous system the RB1 gene was detected at the mRNA and protein levels. The MRPS18-1-3 genes showed different expression patterns, depending on the type of brain tumor. The highest expression of the MRPS18-1 mRNA was detected in glioblastoma samples in both tumor samples and peripheral blood. MRPS18-1 expression was higher in G4 tumors compared to G2. MRPS18-3 was expressed at higher levels in G2 samples and in embryonal tumors. MRPS18-2 was expressed in all samples studied. MRPS18-2 protein signal was detected at high levels in most brain tumors. Assessment of MRPS18-1 gene expression levels in peripheral blood can be used as a potential prognostic marker of tumor progression. Product Description popup.authors Kalman Serhii S.
