Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description1 To investigate the effect of spermine and inhibitors of its catabolism on the viability and molecular biological characteristics of malignant cells of the human prostate in vitro and ex vivo and to determine the potential possibilities of using the most effective combinations of them for antitumor therapy popup.description2 Aldehyde dehydrogenase (ALDH) inhibitors have been found to negatively affect the viability of human prostate cancer (PC) cells in culture. Pargyline (1.65 – 2.4 mM), gossypol (15.4 – 34.0 µM) and disulfiram (19.5 – 25.3 nM) potentiated mainly the apoptotic pathway of death of the more aggressive androgen-resistant cells of the DU-145 line. In the culture of the less malignant androgen-sensitive LNCaP line, the proportion of cells in the state of apoptosis was lower, but ALDH inhibitors inhibited the growth of tumor cells of both lines. During the implementation the stage of project, molecular-biological, biophysical and biochemical parameters were verified, the changes of which correlate with the development of apoptosis and survival of tumor cells. Pargyline, gossypol and disulfiram reduced the expression of cMyc, Bcl-XL and ODC1 genes (at the protein level). Decreased expression of the anti-apoptotic protein Bcl-XL gene was correlated with apoptotic changes in the cells. The obtained data can become the basis for the use of ALDH inhibitors as means of accompanying therapy in the treatment of patients with PC.   Product Description popup.authors Bentrad Veronika V.
