Aim of study: To complete the preclinical dossier of the original molecule by developing a tablet form for clinical trials and to search for new drug candidates among 1,2,4-triazole derivatives through synthesis, in vitro, and in silico studies. Methods: Modern organic synthesis methods were used. Structural analysis involved spectral methods (NMR, IR spectroscopy, mass spectrometry), elemental analysis, high-performance liquid chromatography, and thin-layer chromatography. Biological activity was assessed via validated in vitro tests (cytotoxicity, antioxidant activity, anti-inflammatory properties). In silico methods included molecular docking, QSAR analysis, and ADMET prediction. Preclinical data were systematized and analyzed. Results: Preclinical data on the original molecule were summarized, confirming its therapeutic potential and safety. Proposals for a stable, bioavailable tablet form were developed, completing the preclinical dossier. New 1,2,4-triazole derivatives were synthesized, and their in vitro and in silico studies identified promising drug candidates with potent activity. Practical significance: The results pave the way for a new Ukrainian drug ready for clinical trials. Established synthesis methods and structure-activity relationships provide a foundation for developing original drugs, enhancing Ukraine’s pharmaceutical independence.
