These electrodes, produced using technologies developed at the V. Ye. Lashkaryov Institute of Semiconductor Physics and the Institute of Physics of the National Academy of Sciences of Ukraine by research groups involved in the project, were functionalized with biological receptors—oligonucleotide probes capable of highly selective recognition of specific target DNA sequences. The oligonucleotide-modified electrodes with developed surfaces served as the basis for hybridization DNA sensors, in which the binding of target DNA is detected using electrochemical impedance spectroscopy (EIS). Universal methods for immobilizing oligonucleotide probes on the surfaces of CdS crystals and porous titanium oxide films were developed and optimized. Using impedance spectroscopy, the interactions between immobilized oligonucleotide probes and complementary, partially complementary, and non-complementary nucleic acid sequences were investigated. Strategies for improving the selectivity of the developed DNA sensors were studied, including experiments conducted at elevated temperatures corresponding to the optimal hybridization temperature of the target DNA (i.e., the complementary nucleic acid fragments present in the analyzed samples) with the sensor surface modified by fully complementary oligonucleotide probes. It was demonstrated that the use of these fundamentally new types of electrodes enabled a reduction in the detection limit for oligonucleotides from 1 nM to 0.01 nM, compared to sensor devices based on conventional gold electrodes used for impedance-based signal detection.
