The aim of the work is to analyze genetic and epigenetic changes in tissues and urine from prostate cancer (PC) and bladder cancer (BC); in tissues from breast cancer (BC); to characterize the potential tumor suppressor properties of TAGLN in the baculovirus expression system in vitro. The search is aimed at identifying genes for the diagnosis of malignant neoplasms and the development of gene therapy approaches in model systems. 1. The expression of the RASSF1A genes, long non-coding RNAs ANRASSF1, CCAT1 and GAS5 in tumors and conditionally normal tissues in prostate cancer and bladder cancer was studied. 2. The molecular genetic characteristics of the structure of the BRCA1 gene were determined and hypermethylation of its promoter was detected in breast cancer. 3. The level of expression of the ZFP36 gene and its methylation in breast cancer were studied. 4. The tumor suppressor functions of the TAGLN gene were confirmed and various mechanisms of the influence of this gene on cell death were identified. The results of the studies indicate the possibility of using genetic and epigenetic changes in the studied genes (changes in expression at the RNA and protein levels; methylation status of promoters of protein-coding genes and long non-coding RNA genes) as non-invasive markers for the diagnosis of malignant neoplasms and the development of gene therapy approaches in model systems.
