The aim of the work is to study the expression of a number of long non-coding RNAs in tumor tissues and urine of patients with bladder and prostate cancer and in exosomes of various origins. The search is aimed at identifying genes for diagnosing malignant tumors and developing gene therapy approaches in model systems. A decrease in the relative abundance of RASSF1A gene transcripts and ANRASSF1 antisense dnRNA was found in urine samples of patients with PCM. A strong positive correlation of ANRASSF1 with RASSF1A expression in tumor tissues in PCM was demonstrated. Lower expression levels for ANRASSF1 were found in urine samples of patients with bladder cancer compared to healthy individuals. Changes in the BE of SNHG1 dnRNA in tissues of the corresponding tumors and the relative level of this transcript in urine samples of patients with PCM were found. It was determined that the relative expression of PTEN and SNHG1 dnRNA was higher in tumor tissues of bladder cancer. A negative correlation of SNHG1 dnRNA in urine and treatment was found. A statistically significant negative correlation was found between GAS5 expression in normal tissues and Gleason score. Increased expression of PANDAR in bladder cancer tissues was shown compared to paired normal tissues. A negative correlation was found in patients with prostate cancer between urinary PANDAR levels and PSA levels. An exosome segregation method was developed to study changes in bladder and prostate cancer. The relative level of dnRNA was determined in exosomes isolated from conditioned medium of cell lines, urine and plasma of prostate cancer patients. A number of candidate genes for the regulation of heparin sulfate biosynthesis were identified.
