The purpose of the study: Determination of biophysical characteristics and experimental substantiation of the possibility of using these characteristics as potential therapeutic targets for antimetastatic therapy Research methods and equipment: biochemical methods, cytofluorimetric method using labeled monoclonal antibodies, IR and Raman spectroscopy, confocal and CARS microscopy. Theoretical and practical results: Studies have shown differences in the mechanical properties of the membranes of LLC and LLC/R9 cells depending on their cultivation conditions and under the influence of energy metabolism inhibitors. It has also been shown that deadhesive cells have a more densely packed membrane. The pressure required for complete cell penetration into the capillary is 20% higher than for adhesive cells. A greater spread of the curves of the dependence of the applied pressure on the aspiration radius for adhesive cells was also noted, which can be explained by their higher proliferative heterogeneity. Thanks to the use of an innovative method of drying at the critical point of cells for electron microscopy, it was possible to record fine morphological structures in the form of filopodia and lamellopodia, as well as visualize protein-lipid complexes (lipid droplets) using the CARS microscopy method and correlate morphological and structural rearrangements with spectral features.
