Development and creation of a high-tech vector genetic engineering system based on recombinant baculoviruses, study of vector cells for targeted delivery of target genes to block the progression of malignant tumors in model systems and study the impact of suppressor genes on the development of malignant tumors. popup.description2 The aim of the work was to identify and characterize new potential tumor growth suppressor genes and tumor-associated genes, the expression changes of which accompany the emergence and progression of malignant neoplasms, for the possible diagnosis of tumors of epithelial origin, as well as to develop and create a vector genetic engineering system based on recombinant baculoviruses and study vector cells for the delivery of the studied genes to malignant tumors in model systems. A bioinformatic analysis of open databases was performed to search for potential tumor suppressor genes and tumor-associated genes, genetic disorders of genes associated with drug resistance, and methylation of oncosuppressor gene promoters was also evaluated as a set of possible non-invasive diagnostic markers of prostate cancer. It was shown that an increased level of methylation of the RASSF1A, PTEN and NKX3.1 gene promoters is characteristic of patients with prostate cancer, compared with conditionally healthy individuals. Significant correlations were found between the levels of relative expression of the studied genes and the clinicopathological characteristics of tumor samples and the expression of dozens of tumor- and immune-associated genes. The tumor-suppressor functions of the TAGLN gene were confirmed and various mechanisms of the influence of this gene on cell death were identified. Long non-coding RNAs, in particular ANRASSF1, have been shown to have diagnostic and prognostic potential in bladder cancer. Sets of functional molecular markers (panels) have been developed for certain types of epithelial tumors and diagnostic protocols for the detection and differentiation of oncological diseases based on the created sets have been tested.
