The main barrier on the way to their studies is very low or even absent transcription (so called silent or cryptic genes). It is necessary to generate new systems of controlled expression of these genes to bypass natural regulatory mechanisms limiting their expression. An idea of this work is to develop such expression systems on the basis of primary metabolic genes of the well-studied streptomycete S. albidoflavus J1074. As a result three transcriptional regulatory genes have been selected having the salient prospects as a basis of such expression systems. These are genes XNR_5147 (effector molecule is gluconate, XNR_3053 (valine), XNR_0155 (butyrate). Conditions of their use have been worked out. J1074 strains have been constructed where these genes control the expression of reporter GFP, whose levels can be directly determined.
