During 2024-2025, Ukrainian potato varieties were improved and introduced into in vitro culture: Bazis, Dyvo, Knyahynya, Myroslava, Fotiniya to comply with the European test assortment. Optimization of sterilization modes of plant material of Bazis, Dyvo varieties was carried out to obtain aseptic potato plants in vitro. The most effective was surface disinfection of tuber sprouts with the following reagents: 0.4% silver nitrate solution (for 45-60 seconds) and 0.1% sodium hydroxide solution (for 10 minutes), where 64.7 - 90.2% of the apexes were viable and began development in in vitro culture, and the percentage of plant regeneration was 59.1 - 84.9%. The study of the effect of nutrient media with different compositions on the morphometric parameters of potato plants (varieties Glazurna and Chervona Ruta) in in vitro culture showed that on the 21st day, an increase in plant height and the number of internodes in the 3rd and 4th variants was detected by 1.2-1.8 times compared to the control. The study noted a significant effect on the morphometric parameters of potato plants in the Glazurna and Chervona Ruta varieties when added to the nutrient medium in the 3rd (IOC 1 mg/l + adenine 0.5 mg/l + Xymedon 0.05 ml/l) and 4th (IOC 1 mg/l + adenine 0.5 mg/l + Xymedon 0.01 ml/l) variants. The maximum values ​​of the propagation coefficients were obtained in microplants of the Glazurna variety (3.5 and 3.3) - medium No. 3 and No. 4, respectively, in the Chervona Ruta variety (3.2) - medium No. 3. An increase in the height of potato plants in the studied varieties by 1.2-1.3 times and the number of internodes by 1.6-1.8 times was found when grown in medium No. 3 and No. 4.
