Head Shuba Yaroslav М., Доктор біологічних наук Registration Date 03-02-2026 Organization Bogomolets Institute of Physiology of the National Academy of Sciences of Ukraine popup.description1 The aim of the research is to establish the role of mechanosensitive membrane ion channels from the TRP and PIEZO family in the regulation of vascular and bladder smooth muscle tone (detrusor) and the behavior of stromal cells (connective tissue cells that create the infrastructure of any organ), as well as obtaining experimental facts for the development of methods of photoinduced release of pharmacological substances with channel-specific action using the example of photosensitive nicotine. popup.description2 An integrative analysis of RNA sequencing and differential gene expression data revealed, for the first time, a gradient of differential expression and a functional gradient for inwardly rectifying potassium channels, L-type voltage-gated calcium channels, tubulin, and components of the contractile apparatus between the proximal and distal regions of the rat thoracic aorta. These findings highlight functional heterogeneity of the aorta and reduce experimental variability in vascular studies. Using tensiometry, the contributions of endothelium-derived hyperpolarizing factor (EDHF), nitric oxide (NO), prostanoids, the TRPV4 channel, and non-adrenergic non-cholinergic mechanisms to endothelium-dependent relaxation of rat aortic segments under physiological conditions were quantified, including the relative roles of intermediate- and small-conductance calcium-activated potassium channels in TRPV4-mediated relaxation. In an experimental model of type 2 diabetes mellitus, EDHF contribution was markedly reduced, whereas NO- and prostanoid-dependent components were enhanced. Notably, TRPV4-dependent relaxation was potentiated, accompanied by an increased contribution of small-conductance calcium-activated potassium channels. Furthermore, analysis of calcium-permeable mechanosensitive channels TRPV4 and PIEZO1 in rat urinary bladder detrusor contraction demonstrated that PIEZO1 exhibits a higher activation threshold and steeper stretch sensitivity, being recruited at later stages of bladder filling to limit further volume increase. In contrast, TRPV4 mediates gradual increases in bladder wall stiffness and facilitates muscle fiber adaptation to changes in bladder volume. Product Description popup.authors Yaroslav М.
