In case of untimely termination of pregnancy, preeclampsia turns into eclampsia - convulsions, which often end in the death of mother and child. Placental dysfunction plays a leading role in the occurrence of preeclampsia. The aim of the project is to conduct a systematic analysis of the transcriptome, proteome and metabolite in the human placenta under physiological conditions of pregnancy, preeclampsia and in women smokers, in whom the incidence of preeclampsia is twice lower than in women who do not smoke. Identifying the factors that contribute to and support the disease and that prevent its occurrence is one of the features of this project. To achieve the main goal of the project, two approaches will be used - bioinformatics analysis of publicly available data and experimental studies aimed at validating the results of bioinformatics analysis, search for new markers of the pathological process and factors that contribute to seizures. popup.description2 Objective. To analyze differentially expressed genes in the human placenta between the first and second trimesters of physiological pregnancy according to previously defined clusters. Results. Clustering of differentially expressed genes in the human placenta between the first and second trimesters of physiological pregnancy revealed four functional clusters. Within the “Immune processes” cluster, genes associated with the following functional categories were identified: pattern recognition receptors and the complement system, neutralization of potential pathogens, migration of immune cells, transplacental IgG transport, and immune tolerance. The cluster “Morphogens, extracellular matrix, and angiogenesis” contained the largest gene categories, which in descending order included: G protein–coupled receptor signaling; organization of the extracellular matrix (ECM); positive and negative regulation of cell proliferation; cell adhesion; and positive regulation of angiogenesis and apoptosis. These categories predominantly reflect biological processes related to angiogenesis and the function of various components of the extracellular matrix, which determined the designation of this cluster. At the end of gestational weeks 10–12, maternal blood enters the intervillous space, resulting in a transition of placental nutrition from histiotrophic to hematotrophic. This transition is supported by the development of the placental vascular system, which occurs in parallel with the growth and functional maturation of the organ. Conclusions. During the transition from the first to the second trimester of gestation, morphogenetic processes continue in the human placenta with the active involvement of morphogens, whose effects are balanced by the action of activators and inhibitors.
