In case of untimely termination of pregnancy, preeclampsia turns into eclampsia - convulsions, which often end in the death of mother and child. Placental dysfunction plays a leading role in the occurrence of preeclampsia. The aim of the project is to conduct a systematic analysis of the transcriptome, proteome and metabolite in the human placenta under physiological conditions of pregnancy, preeclampsia and in women smokers, in whom the incidence of preeclampsia is twice lower than in women who do not smoke. Identifying the factors that contribute to and support the disease and that prevent its occurrence is one of the features of this project. To achieve the main goal of the project, two approaches will be used - bioinformatics analysis of publicly available data and experimental studies aimed at validating the results of bioinformatics analysis, search for new markers of the pathological process and factors that contribute to seizures. popup.description2 Objective. To identify and visualize enriched biological processes and to further analyze differentially expressed genes involved in immune processes in the human placenta between the second and first trimesters of pregnancy. **Results.** Based on data on differential gene expression in the human placenta between the first and second trimesters of pregnancy, a volcano plot was constructed, and the top 20 genes were characterized. Among them, four are involved in hormonal processes: **HSD11B1**, corticosteroid 11-beta-dehydrogenase type 1, which reversibly catalyzes the conversion of cortisone to active cortisol; **HSD17B2**, Hydroxysteroid 17-Beta Dehydrogenase 2, catalyzes the NAD-dependent oxidation of highly active 17β-hydroxysteroids such as estradiol (E2), testosterone (T), and dihydrotestosterone (DHT) to their less active forms, thereby regulating the biological activity of these steroids; **HSD3B2**, Hydroxy-Delta-5-Steroid Dehydrogenase, 3 Beta- And Steroid Delta-Isomerase 2, a bifunctional enzyme that catalyzes the oxidation of Δ5-ene-3β-hydroxy steroids and keto steroids. The 3β-HSD enzymatic system plays a critical role in the biosynthesis of all classes of steroid hormones. In the placenta, HSD3B1 is considered to play a key role in progesterone synthesis. SCGB2A1 (Mammaglobin B) can bind androgens and other steroids, and may also bind estramustine, a chemotherapeutic agent used for prostate cancer treatment. It may be regulated by steroid hormone transcription and belongs to the secretoglobin family. Eight genes are involved in immune processes, including neutralization of bacteria and damaged cells (C7, DEFB1), placental tissue tolerance to the allogeneic fetus (FCGR2B, RAET1E, GPC6), and migration of immune cells from the bloodstream to sites of inflammation (EPCAM, GPC6, JAM2).
