Head Telehieiev Henadii D., Доктор біологічних наук Registration Date 24-02-2026 Organization Institute of Molecular Biology and Genetics of NAS of Ukraine popup.description1 The aim of the work is to study the factors that determine the onset, tumor progression and the formation of chemoradioresistance in patients with myeloproliferative human neoplasms (MPN) and in the development of diagnostic and possibly therapeutic approaches on this basis. popup.description2 It has been established that inhibition of USP1 activity using trifluoperazine and pimozide and influencing GLG1 expression levels using memantine causes a dose-dependent decrease in the viability of DU145/RR and LNCap PP/RR cells. A cytoplasmic localisation atypical for the nuclear protein USP1 was detected in radio-resistant DU145/RR and LNCap PP/RR cells. The DU145 cell line is more sensitive to inhibitors than the radioresistant DU145/RR cell line, which is able to neutralise oxidative stress more effectively and compensate for damage. RR requires higher concentrations for ROS to reach toxic levels. Inhibition of USP1 and modulation of GLG1 expression lead to destabilisation of key cellular regulatory mechanisms, disruption of homeostasis and vital functions, with the nature and strength of the response to trifluoperazine and memantine depending on the phenotypic and metabolic characteristics of individual prostate cancer lines. A reverse correlation was demonstrated between the expression of leukaemic stem cell marker genes SNAIL, OCT4 and PLAP and myeloid differentiation marker genes C/EBPα, TNAP, and E-cadherin in K562 cells under the action of alpha-tocopherol or G-CSF, which indicates the targeted activation of the molecular pattern of myeloid differentiation of CML blast cells under suppression of the leukaemic stem cell phenotype.
