Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description1 To study the influence of the metabolic microenvironment, associated with the different stages of the metastatic cascade, on the survival rate and energy metabolism parameters of metastatically active cells and to reveal the role of LDH in the ability of these cells to adapt under variable survival conditions popup.description2 It has been proven that energy metabolism inhibitors, unlike cisplatin, exhibit significantly higher cytotoxicity against deadhesive LLC cells compared to adhesive ones. It has been established that under deadhesive growth, lactate dehydrogenase activity increases significantly, especially in cells with lower metastatic potential, which indicates intense compensatory activation of glycolysis as a survival mechanism under the threat of anoikis. In the passive metastasis model, it has been demonstrated that the deadhesive LLC cells exhibit higher metastatic potential. Pretreatment of adhesive LLC cells with oxamate and metformin significantly reduces the number of lung metastases. Oxamate at low doses does not affect tumor growth and metastasis of LLC, while high doses exhibit antitumor and antimetastatic effects. Thus, it has been shown for the first time that oxamate and metformin exhibit pronounced cytotoxicity against circulating metastatic Lewis lung carcinoma cells. Pretreatment of adhesive LLC cells with oxamate in combination with metformin synergistically increases their sensitivity to anoikis, indicating potential antimetastatic activity. In vivo studies have demonstrated that preincubation of tumor cells with oxamate, in combination with metformin, inhibits their metastatic potential, resulting in a significantly higher percentage of animals without lung metastases.
