Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description1 To investigate the effect of spermine and inhibitors of its catabolism on the viability and molecular biological characteristics of malignant cells of the human prostate in vitro and ex vivo and to determine the potential possibilities of using the most effective combinations of them for antitumor therapy popup.description2 For the first time, the synergistic effect of the cytotoxic and proapoptotic action of spermine (Spn) and silibinin (Sil) on prostate cancer cells, which is realized through the mechanism of "tandem aldehyde shock", has been shown. In particular, the combined use of Sil and Spn led to the blocking of ALDH-dependent cellular detoxification pathways. At the same time, the indicated combination had a quantitatively opposite effect on the proliferative activity of the studied cell lines: the less malignant LNCaP line showed a higher sensitivity to the action of Sil, while the more aggressive DU-145 line showed a higher sensitivity to Spn. The obtained data form the fundamental basis for the development of schemes for the use of these drugs as adjunctive therapy in the treatment of patients with prostate cancer.
