Subject of this investigation - heme metabolism and some parameters of antioxi-dative-prooxidative status of rat's liver, kidney and spleen under sodium nitroprusside administration in different doses. The heme oxygenase, tryptophan-2,3-dioxygenase, 5- aminolevulinic acid synthase, su-peroxide dismutase, catalase activities, cytochrome P-450, total heme content and the level of spontaneous and ascorbate-induced LPO and MDA and reduce glutathione content in liver, kidney and spleen of rats and absorbtion spectrum of blood serum in Soret region at 2 and 24 h after sodium nitroprusside administration. The Wistar rats were used in the work. Sodium nitroprusside in chosen doses (10 mg/ 100 g and 100 mg/ 100 g) doesn't make influence on level of spontaneous and ascorbate-induced LPO and MDA and reduce glutathione content in liver, kidney, spleen and blood serum but causes certain changes of antioxidant enzyme - superoxide dismutase and catalase ac-tivity. Heme oxygenase activity in liver decrease under sodium nitroprussid in dose 10 mg/ 100 g and in kidney - under sodium nitroprusside in dose 100 mg/ 100 g. Incuba-tion of liver homogenate with sodium nitroprusside causes a heme oxygenase activity decreasing.Total heme content falls in liver after addition of a NO donor. 5-ALAS ac-tivity in liver was elevated under sodium nitroprusside treatment, but content of free heme does not essencially change. Cytochrome P450 level in liver falls after sodium ni-troprusside administration in vivo and in experiences in vitro. Thereby, date obtained suggested that NO donor (sodium nitroprusside) in chosen doses does not make an essential influence on antioxidative-prooxidative homeostase in organs investigated. The decrease of total heme content, some hemoprotein level and heme oxygenase activity revealed is probably caused by heme nitrosilation by NO radi-cals.
