Technological regimes for obtaining of mature dendritic cells from monocytes of peripheral blood and from excudate in the patients with advanced ovarian cancer and from spleens of mice, methods for obtaining of lysates of tumor cells (LTC), tumor antigens (TA) and bioengineering technology of mechanomodification (m/m) of TA have been developed. Using phenotyping, functional tests and cytomorphological examinations it has been proved, that using of the developed methods it is possible to obtain of mature DC. The most adivible terms for obtaining of DC with taking into account the number and terms of PCT courses in ovarian cancer patients are determined. Anticancer vaccines of various constructions on the basis of DC, LTC, TA and m/m TA have been created and their anticancer and antimetastaticeffect dependently on the ways of their administration was studied. It has been shown that the most effective are DC loaded with TA at intracutaneous administration as well as DC loaded with m/m TA at i/v administration. It has been detected that increase of the number of administrations of DC-vaccines promotes potentiation of its antimetastatic effect. Realization of antitumor and antimetastatic effect of DC-vaccines is at the expense of normalizing influence on lymphoid organs and potentiation of functional activity of cells which stipulate state of natural antitumor resistance of organism: cytotoxic activity of natural killer cells and amount and phagocytic activity of phagocyting splenocytes.
