Technological regimen for obtaining of dendritic cells from peripheral blood leukapheresis monocytes in the patients with cancer have been developed. Using phenotyping, functional tests and cytomorphological examinations it has been shown, that maturity of obtained DC depend on concentration of tumor antigen and route of their administration. Realization of antitumor and antimetastatic effect of DC-vaccines is at the expense of stimulating influence on lymphoid tissue of recipient. DC administration results in increase of amount of lymphoblasts in thymus, immunoblasts, plasmacytoid cells, CD3+,CD4+,CD8+ - lymphocytes in spleen, absolute amount of lymphocytes, CD8+ - lymphocytes and large granular lymphocytes in peripheral blood, as well as essential increase of cytotoxic activity of splenocytes. Insrease of CD4+/CD8+- lymphocytes correlation in spleen and decrease of functional activity of DC in vitro in mixed leukocyte reaction with allogeneic lymphocytes is followed by worse antimetastatic efficiency of DC, loaded with lyophilized tumor cells (LTC) or mechanomodification (m/m) LTC.
