Head Pilinskaya Maria, Доктор медичних наук Registration Date 17-01-2007 Organization Research Centre for Radiation Medicine popup.description2 Object: the blood lymphocytes of the nonirradiated persons. Goal: To develop the model system for revelation of latent radiation induced chromosome instability in somatic human cells with testing mutagenic loading with bleomycin in vitro (to adapt G2 bleomycin sensitivity assay). Methods: cytogenetic analysis of human peripheral blood lymphocytes. The model test-system for the investigation of hidden chromosomal instability in somatic human cells with the help of bleomycin mutagenic exposure in vitro had been elaborated. Cytogenetic activity of four bleomycin concentrations in the culture of human peripheral blood lymphocytes from noniradiated persons had been checked and optimal concentrations of the drug (0,05 and 5,0 mkg/ml) suitable for the evaluation of human somatic chromosomes sensitivity to mutagenic exposure in vitro had been selected. It had been shown that the main criterion of human chromosome sensitivity to testing bleomycin exposure it worth while to consider the total frequency of chromosome aberrations (but not aberrant cells). It had been estimated that for the identification of susceptible persons with increased chromosomal sensitivity to bleomycin exposure both concentrations of the drug necessary to use with the help of which three hypersensitive individuals had been determined. This investigation had been allowed to adopt "G2-bleomycin sensitivity assay" for revealing of the individual radiation-inducted hidden chromosome instability in somatic human cells.
