Head Chumak Anatoliy Andreevich, Доктор медичних наук Registration Date 05-02-2007 Organization Research Centre for Radiation Medicine popup.description2 The object of study - 117 B-cell chronic lymphoid leukemia (B-CLL) patients. The aim of study is to determinate VDJ rearrangement of B-CLL patients, who t[hjstd to ionizing radiation due to Chornobyl NPP catastrophe and correlation between genetic features of leukemic cells and TORCH-infection. Methods of study are polymerase chain reaction with following sequencing of PCR products, immunoenzyme assay, statistical methods. The structure of VDJ rearrangement of heavy chain immunoglobulin genes in chronic lymphocytic leukemia (CLL) patients who were exposed to ionizing radiation habe been analyzed. It was found the prevalence of cases with unmutated VH3 and VH4 genes (negative prognostic factor), the absent of cases with prognostic favourable VH3-23 gene, increased number of cases with unmutated VH3-09 gene with worse course of disease. Some dependence between dose of irradiation and increased number of unmutated VH3 and VH4 genes was revealed. The homology of nucleotide sequences of VH1 genes (7 of 9 VH1+ patients) with early described was found. It suggest about influence of the same antigen stimules in different regions for CLL induction. High seropositivity of patients for persistence infections and increasing number of VH3 genes in CMV and Toxoplasma gondii seropositive patients were revealed, that indicate on the possibility of stimulation of certain B-lymphocyte's clones by viral and bacterial antigens.
