It has been revealed that there is a direct dependence of antitumor effect of dendritic cells (DC) on a dose of tumor antigen (TA) used for their loading. In study of antimetastatic activity of DC such dependence was not observed. Dependence of antitumor effect of DC on their amount and ways of infusion is determined. It is proved that in animals with tumors their ability to migrate is significantly decreased independently of the way of their administration as compared with intact animals. The highest migration activity of DC to lymphoid organs occurs in intravenous way of administration, the lowest – in subcutaneous. Use of TA with increased immunogenic properties (lysate of tumor cells treated by lectines Bacillus subtilis B-7025) for loading of DC potentiates their antimetastatic effect. It has been detected that combination use of DC-vaccines with preparation “Ukrain” is significantly effective as for inhibition of development of metastases of melanoma B6 to lung of mice as compared with their separate use. Dependence of phenotypic properties of DC of monocytic origin on dose of TA for loading is studied. It has been found that “Ukrain” stimulates maturation of DC with maximal effect at concentration of 0,6 ?g/ml, which is a therapeutic one, in tissues of the patients after use of preparation. A protocol of DC-vaccine therapy in complex treatment of patients with ovarian, lung cancer and melanoma has been used. It has been proved that it is possible to obtain mature DC suitable for use as antitumor vaccines in patients with various forms of malignancies.
