It was shown that the spherical Au nanoparticles are sensitive to large overlayer thicknesses; however, the shift of the localized surface plasmon resonance peak (LSPR response) in the light extinction spectrum for thicker overlayer grows slower with an increase in overlayer thickness compared to the same increase in that of a thinner overlayer. It was revealed that the upper limit of sensitivity to biomolecular cover is smaller but comparable to that of the conventional surface plasmon resonance (SPR) technique at equal nanoparticle diameter and gold film thickness in SPR. It was demonstrated that LSPR response for small nanoparticles (with a radius up to 5 nm) could be positive or negative depending on the nanoparticle size and biomolecular cover thickness. It was also found out that contribution of scattering into total extinction depends on the ratio between nanoparticle and overlayer sizes, namely, scattering increases and could lead to negative LSPR response upon the increase of nanoparticle radius and cover thickness. Equations for the number of globular biomolecules and their surface concentration for saturated biomolecular monolayer on the surface of the nanoparticle were derived for further use in description of biomolecular cover by means of effective medium theory.
