Nalyvaiko Description The dissertation is dedicated to conducting epizootiological monitoring of avulaviruses in their natural reservoir, identifying the characteristics of their circulation in natural hosts of various species, and improving the serological diagnosis of avulaviruses as part of the epizootiological monitoring system in Ukraine. A comprehensive approach to diagnostics ensures the detection of the circulation of new epizootic isolates and the introduction of new serotypes into new geographical regions. New scientific data have been obtained, expanding the existing information on the ecology of avulaviruses in birds in their natural reservoir, as well as their role in maintaining natural circulation. Epizootiological monitoring of wild birds belonging to various ecological groups, including 62 species from 8 orders, has been conducted to isolate avulaviruses of different serotypes. This has enabled us to confirm their wide circulation. Most of the viruses were first isolated in Ukraine from wild birds. Specifically, 26 isolates have been obtained, from which 6 different serotypes of avulaviruses have been identified, confirming the serotypic diversity of avulaviruses and cross-reactions in 27 isolates. Research was conducted for the first time to study certain biological properties of avulaviruses, namely AaV 1, AaV 4, AaV 6, and AaV 7 serotypes. The potential of using them as components for diagnostic studies has been demonstrated, particularly for the accumulation and extraction of positive serum which was subsequently used for the identification of field isolates for internal laboratory research. For the first time in Ukraine, a technological protocol for the production and quality control of the “Kit for detection of antibodies to Newcastle disease virus in hemagglutination inhibition test” has been developed. As a result of interlaboratory and interdepartmental commission tests, the test system was registered in Ukraine and obtained a registration certificate. The practical significance of the thesis lies in the development of technological approaches for obtaining inactivated antigens for hemagglutination inhibition test to generate positive sera. These positive sera are used in the identification of field isolates for in-house laboratory use. Additionally, a domestic test kit has been developed for detecting antibodies in blood sera and egg yolk extracts against Newcastle disease virus in hemagglutination inhibition test. This system is utilized for monitoring all bird species to determine the presence and level of antibodies in infected (wild birds) or vaccinated (agricultural) birds.
