So in the sensorimotor cortex and hippocampus CA1-zone occurred both structural and quantitative changes in the cells. Growing share of hyperchromic neurons as well as macro- and miсrogliosis were observed. The loss of neurons in the cortex after TBI in adult animals was higher than in the hippocampus and in old - vice versa. However, in the cortex and in the hippocampus of older rats after TBI neurons approach the critical point of the same - 67% of the amount of intact adult. After TBI the degree of gliosis was the same both as in adults, and the old; the number of endothelial cells (EC) increased greater in adults than in older ones. Apoptotic index (AI) after TBI increased in adults more than in the old, but in the old the baseline of AI was significantly higher than that in adults. Gene therapy by means of liposomes with a plasmid carrying the human ApoE3 gene (GT) has reduced the neuronal loss that was dependent on age and brain areas: decreasing the loss in the cortex of 6% in adult and 2 times in old hippocampus - 8% in adult and 13% in old. GT also reduced macrogliosis: in the cortex - 3.2 times in adults and 1.6 times in old hippocampus - 12% in adult and 2 times in old. Thus, the GT did not change the AI which was determined in 10 days after TBI. The therapeutic effect of GT is more pronounced in older animals. GT did not significantly affect the level of microgliosis and the number of EC. Thus, a decrease of reactive gliosis and neuronal loss in the cerebral cortex and hippocampus of adult and old rats which were treated with GT indicates significant restorative effect of GT and its promising after TBI.
