Our strategy is based on the exploitation of structural and functional differences between pathogen prolyl-tRNA synthetase (ProRS) and its human homologue. The search will use the combination of several approaches: X-ray crystallography structure and molecular docking of compounds followed by a target-specific screening by biochemical in vitro reactions. By the molecular docking methods was selected 48 chemical compounds as perspective inhibitors of enzyme and carrying out biochemical essays in vitro. 180 mg of E. faecalis ProRS was isolate and purified, and optimal functioning conditions in vitro was find. Inhibiting activity of 48 compounds was study and 8 the most perspective compounds that inhibit activity of ProRSEF by 31-37% was select for detailed study and use for repeated molecular docking cycle. It was analysed and submitted to the database 249 new genomic sequences encompassing DIS, SD and Psi hairpins. New variants of these hairpins have been found and our recently published hypotetical schemes of their evolution have been renewed. The PBS domain structure in 188 HIV-1 isolates of different groups and subtypes have been analysed. Certain combinations of base changes in this domain were demonstrated to be highly specific to virus subtype and modulate specifically the domain's secodary structure.
