Producers of landomycin E, landomycin A, urdamycin A (Streptomyces globisporus 1912, S. cyanogenus S136, S. fradiae Tu2717, respectively) and their derivatives constructed in course of this work, were utilized. Aim of the work - experimental verification of the potential of structural and regulatory genes for angucycline biosynthesis to perform combinatorial biosynthesis and detection of novel bioactive compounds. Methods of investigations: cloning and sequencing of genes, conjugal matings, recombinant DNA construction, DNA hybridization, PCR, thin layer and high performance liquid chromatographies, isolation and purification of proteins, microscopy. Set of recombinant actinomycete strains were designed overexpressing late genes for angucycline biosynthesis. Glycosylated derivatives of prejadomycin were isolated for the first time. True acceptor substrate for UrdGT2 has been identified. The organization and role of Streptomyces globisporus 1912 and S. cyanogenus S136 genes for landomycin glycosylation and export have been studied. Transporter genes lndJ and lndW conferring streptomycetes to landomycin resistance were examined. Sequencing and analysis of regulatory genes lanI, prx1, lndY, lndYR were completed. LndGT4 overexpression system has been developed. In S. cyanogenus, expresion of lanJlanZ1lanGT3lanZ2lanGT4 genes is represed by LanK. Landomycins with long glycoside chains (no less than 3 sugars) block LanK. Gene lanK was used for biosensor-guided identification of known and novel members of landomycin family with extended glycoside chains.On the basis of these results, a patent of Ukraine has been filed (registrated 8.01.2008, Na200800261). The reuslts of research work can be used during construction, selection and screening of actinomycetes-producers of antibiotics and scholar process of preparation of geneticists and biotechnologists.
