It was shown that under normal conditions -opioid receptor agonists inhibit P2X2/3-mediated currents, activated by ATP, by 50% at the average via G-protein dependent mechanisms. Co-culturing of neurons with mice fibrosarcoma cells (NCTC 2472 cell line) for 2-5 days leads to significant decrease of endomorphin-1 inhibitory action on ATP-activated currents with slow desensitization kinetics. At the same time relative number of neurons with slowly desensitizing currents also increases. Besides, we studied the mechanisms of opioid modulatory influence on P-type voltage-gated calcium channels in Purkinje neurons. Our data indicate that opioids act via or receptors of a neuron, altering neuronal excitability by bimodal regulation of P-type voltage-gated calcium channels.
