The purpose: to receive the transfer factor from lymphocytes of animals, immunized by cells of xenogeneic tumours, and to determine it tumour-specific action. Methods of research - immunological, cytofluorimetrical, a method of cell culture, statistical. The equipment: centrifuges ОPN-3, 8, Т 23; a microscope; a photometer; flowing cytometer FACScan; the automatic sampleres; termostates. Methodical approaches of reception from lymphocytes immunized animals of the transfer factor (TF) specific to cells of xenogeneic tumour were optimized. The technology is developed and samples TF of rats, immunized by cells or a tissue Lewis lung carcinoma (LLC) mice C57BL/6 were received. Antineoplastic and antimetastatic TF effects of xenogeneic origins were investigated on the model of experimental tumoral growth. On the model of passive metastatic disease LLC at mice C57BL/6 it is established, that introduction of samples TF received on a peak of the immune answer after immunization of rats by cells or transplantation into spleen the LLC fragment (TF14-І, TF14-ІІ) and during formation immunological memories after intraperitoneal immunization of rats by cells LLC (TF60-І), causes immune protection of an organism against development of metastasises to what testifies expressed antimetastatic effect which is shown by reduction of frequency metastatic disease, decrease of quantity and volume of lung metastases, a high index of tumour dissemination inhibition. The samples of xenogeneic TF (TF14-І, TF14-ІІ, TF60-І) also show antimetastatic action at their application with the purpose of biotherapy of mice C57BL/6 with LLC. Long introduction of xenogeneic TF (once a week, 5 injections) provides reduction of frequency metastatic disease, total and volume of metastasises LLC. Except for that these samples TF, as against nonspecific TF, increase by 28-42 % life expectancy at regular introduction to mice C57BL/6 with LLC. Adjuvant immunotherapy TF14-І or TF14-ІІ promotes the prevention metastatic disease (accordingly in 50 % and 23 % of animals) or reduction of quantity and volume of lung metastases at mice C57BL/6 with the removed tumour. These samples induce in an organism of mice antineoplastic immune reactions concerning LLC antigenes that is shown іn vіtro active proliferation of lymphocytes at the presence of LLC cells. At іn vіvo research it was established, that samples TF14-І, TF14-ІІ and TF60-І are capable to transfer delayed type hypersensitivity on antigenes LLC to the recipient. In system іn vіtro it is shown, that TF samples received after immunization of rats by cells or transplantation of fragment LLC into spleen, induce more expressed answer of lymphocytes on gomological LLC cells in the mixed culture in comparison with action nonspecific TF of xenogeneic origins. Degree of introduction - methodical approaches on reception of tumour-specific TF of xenogeneic origins are optimized; the developed methodology is introduced in research laboratory of clinical immunology of National cancer institute. Field of application - development concerns to medicine, in particular oncology and pharmacology, and can be applied in manufacturing techniques of immunomodulators which are used at treatment of malignant patients.
