The aim of the work: to develop the experimental sample of compact analyser on the base of amperometric enzymatic biosensors for analysis of wines and control their quality. The aim of the work, part 3 (ICB, NAS of Ukraine): isolation of preparative quantities of enzymes: alcohol oxidase from Hansenula polymorpha and glycerol oxidase from Bоtrytis allii. The aim for the 2009 year stage (part 3): to compare the data of biosensoric and referent methods for determination of ethanol, lactate, glycerol and glucose in wines and wine's products. Methods of investigations: microbial, chemical, physico-chemical, analytical, enzymatic. Results of the work: optimal conditions for microbial strains overproducing alcohol oxidase and glycerol oxidase were investigated; the cells with high yield of the enzymes were obtained: preparative isolation, purification, stabilization were made and physico-chemical characteristics of enzymes (alcohol oxidase and glycerol oxidase) as bioselective elements of biosensors were investigated; the determination of glucose, ethanol and glycerol content in model solutions and real wine's samples by standard methods was done; the new enzymatic photometric method for lactate analysis based on flavocytochrome b2 of the yeast H. polymorpha was developed and tested on the real samples of wines; the comparison of the data obtained by using different analytical methods (enzymatic and biosensoric) for ethanol, lactate, glycerol and glucose assay in wines was done.
