Ovcharenko Institute of biocolloidal Chemistry NAS of Ukraine popup.description2 During Project performance it was developed methods for synthesis and are created eight new nanoforms on a basis of gold, platinum nanoparticles (NP) and nanoprisms (Npr): nanobioconjugates-BC (AuPS and PtPS) with polysaccharides (PS) of microalgae Chlorella vulgaris; nanocomposites-NC gold NP - doxorubicin(DOX)- polysaccharides (Au-DOX-PS, the form - Dox-inside) and NP gold-polysaccharides-doxorubicin (Au-PS-DOX, the form - Dox-outside); platinum-doxorubicin-polysaccharides (Pt-DOX-PS -Dox-inside), platinum-polysaccharides-doxorubicin (Pt-PS-DOX -Dox-outside); magnetite-albumine-gold, magnetite-albumine-platinum. Polysaccharides have been used taking into account known data about PS antineoplastic properties of microalga, in particular Chlorella. Stability/aggregation of synthesised BC and NC with protective adsorption layer of Chlorella vulgaris PS in a normal saline solution and in a blood plasma was investigated. It was shown, that optical density of the conforming admixtures does not variate within 2 hours in a blood plasma and over a period of half a year - in a normal saline solution. Such researches are directed on the decision of a problem of storage of such drugs and a thrombosis exception after intravenous injection. In study in vitro cytotoxic action of synthesised nanoforms has been investigated and proved with cells of an ovary cancer line А2780 and melanoma А375. It was shown that an index of toxicity IC50 of synthesised nanoparticles, BС and NC are in an interval of concentrations on metals 0,05-0,2 mM and in the rank: PtPS <AuPS <PtCl2PS <Pt <Au. Efficiency of cytotoxic influence of NC Au-DOX-PS and Au-PS-DOX was studied for the cells A2780S under the influence of cells contact with such NC depending on their concentration and contact time. It is shown, that such efficiency corresponds to dependence: Au-DOX-PS> Au-PS-DOX. Efficiency of influence NC Pt-DOX-PS and Pt-PS-DOX on tumor cells A2780S corresponds to dependence: Pt-DOX-PS> Pt-PS-DOX. Concentration of toxic doxorubicin decreases in comparison with its known medical dose. The complex of researches of eficiency of BC AuPS and PtPS aggregation with tumor cells, including methods for determination of colloid sorption, kinetics of heterocoagulation and kinetics of cytotoxic effect is offered. It was shown, that with a surface of one cell A2780S aggregate 1,2·103-2,5·104 particles of AuPS (during the change of AuPS concentration СAu <0,023?0,185> mM) and for PtPS - 6,9·104-1·106 particles per cell (СPt <0,185-0,65> mM). It is shown, that characteristic time of aggregation cells-BC particles is for BC AuPS 10-15 minutes, for BC PtPS - 20-25 minutes During 1 hour the cytotoxic effect of BC AuPS continuously increases, and for BC PtPS cytotoxic effect reaches saturations in the first 25 minutes. In experiences in vivo it has been shown medical effect of nanogold in form of BC AuPS and NC Au-DOX-PS, Au-PS-DOX on a cancer tumour of sarcoma S180 (test animals - mice). At half of mices the sarcoma tumour has disappeared in 2-3 weeks after one i.v. injection of such drugs in a normal saline solution. From known literary data such medical effect of BC and NC on the basis of gold nanoparticles is received for the first time. Till now is widely know the result of bunch of prof. El-Sayed'a (the USA, 2008) (where weaker compared with ours the medical effect) has been received only at use in a complex from NP of gold with a method of laser photothermal therapy. Regular histological researches of changes of morphology of a liver and nephroses of tested mice under the influence of entered i.v. BC AuPS (doses from 0,54 to 47,6 mgAu/kg of weight) are carried out or the first time. Appreciable changes are observed at a dose above 3,0 mgAu/kg of animal weight.
