A number of interactions between components of PI3-dependent signaling pathway namely, S6K1 kinase and TSC1/2 complex with other proteins have been identified by yeast two-hybrid system. Interaction of S6K1 and TSC1/2 with some of the identified proteins - CK2, ubiqutinligase Roc1 and PP5 phosphatase have been studied in more details. Novel mechanism of CK2 mediated regulation of subcellular localization of S6K1 and its stability through the mediated by Roc1 ubiqutine dependent proteasomal degradation have been discovered. Additional mechanism of negative regulation of TSC1/2 complex involving PP5 phosphatase have been revealed. New izoform of mTOR kinase - mTOR? have been identified and characterized. Oncogenic properties of mTOR? have been demonstrated. New mechanism of activity regulation of metabolic enzyme CoA synthase by phosphates Shp2 defosforylation have been proposed and oncogenic properties of CoA synthase have been demonstrated. Phosphate transporter Napi2b have been identified as a novel marker of epithelial ovarian cancer and its expression in tumors of different hystotype have been studied. Conditions for 3D cultivation of MCF7 cell line and acinus of breast tumors have been developed.
