It was found that with age in mice streptozotocin-induced diabetes weakened. Thus, the increase in blood glucose concentration, glycosylated hemoglobin and lipid peroxidation products, as well as reducing body weight and lymphoid organs were observed in a larger number of cases and were more distinct in adult mice, compared with older animals. In the investigated tissues of diabetic mice were found some age differences in gene expression of pro-and anti-inflammatory cytokines. In particular, only in animals of younger age group with diabetes noted increased expression of proinflammatory cytokine TNF-a in bone marrow cells, while only in old mice - induced increase of bacterial lipopolysaccharide (LPS) expression level of TNF-a and IL-1b in the aorta and weaken expression of inflammatory cytokine IL-10 in spleen macrophages. Triggered diabetes complications of the bone and cardiovascular systems in mice of different age also manifested in different ways. Thus, in adult animals bone mineral density (BMD) decreased in diabetes and positively correlated with the expression level of TNF-a and IL-1a in macrophages, whereas in the old - practically not changed, and negatively correlated with expression of TNF-a and IL-1b in macrophages. If in adult mice, diabetic status was characterized by increment of а1-agonist vasoconstriction and the development of tolerance to LPS, in the old - the weakening of endothelial vasodylatation, especially against the background of LPS and blockade of endogenous prostanoids. Application of preparation Corvitin did some positive impact on the manifestations of experimental diabetes in mice. In particular, the course introduction Corvitin dose of 13 mg / kg depressed increased due to diabetes gene expression of TNF-a in bone marrow and restore BMD to levels typical of healthy animals. Introduction Corvitin dose of 130 mg / kg impaired expression of hyperglycemia, oxidative stress and autoimmune destruction of pancreatic islets in diabetes. In old mice with diabetes use Corvitin in vitro prevented LPS-induced violation of relaxation of the vascular wall to acetylcholine. Thus, with aging in mice may impair symptoms of streptozotocin diabetes and changing manifestations of diabetes-associated pathologies. Features of development of diabetic complications in animals of different ages may be caused by age differences in the regulation of inflammation process by pro-and anti-inflammatory cytokines. The drug Corvitin can weaken manifestations of experimental diabetes and prevent some of its complications.
