The purpose - To determine features of shifts in immune system of patients skin melanoma under influence interferon therapy for forecasting efficiency of the combined treatment. Methods of research - immunological, flow cytometry, a method of cell culture, statistical. The equipment: centrifuges ОPN-3; a microscope; flowing cytometer FACScan; the automatic sampleres; termostates; the СО2-incubator. Clinical and immunological investigation of 26 patients with the histological confirmed diagnosis primarily-located skin melanoma (PLМ) of a ІВ-ІІС stage in dynamics of the combined treatment with application of different interferon therapy regimes has been studied. It is established, that consecutive application recombinante preparations IFN-gamma and IFN-alpha2b in аdjuvant regimen promotes preservation of quantity of leukocytes within the limits of physiological norm and prevents development absolute neutropenia and lymphocytopenia which occurrence is not possible to avoid during аdjuvant IFN- alpha 2b therapy. Both interferon therapy regimes essentially do not change population structure peripheral blood lymphocytes, but differently influence contents Тrеg and В1-cells at PLМ patients. Application IFN- alpha2b in аdjuvant regimen promotes decrease in absolute quantity Тrеg and increase in the relative contents of В1-cells that can be the precondition of autoimmune changes development; there is an increase of relative contents CD95+-lymphocytes, that can explain development absolute lymphocytopenia at patients of the given group. Аdjuvant interferon therapy with combined application IFN- gamma and IFN- alpha2b results in increase in relative quantity Тrеg in peripheral blood at PLМ patients and reduction of relative amount activated HLA-DR+-lymphocytes and it is no influenced in contents CD95+-lymphocytes. Application IFN- alpha2b in аdjuvant regimen results in reduction mitogen-induced lymphocytes proliferation (through 3 month from the beginning interferon therapy), and consecutive application IFN- gamma and IFN- alpha2b is accompanied by its significant increase not only compared with an initial level, but also with parameters at practically healthy people. Any of circuits interferon therapy in аdjuvant regimen essentially does not influence on cytotoxic blood lymphocytes activity of PLМ patients. The received results have preliminary character as at the given investigation phase there is no opportunity to estimate efficiency of the applied circuits interferon therapy, thus it is necessary to continue a set of a clinical material and to continue supervision over patients at a stage after the combined treatment. Comparison of results of influence of IFNs on immune system of patients with efficiency of the combined treatment will enable to make more substantiated conclusions.
