It will be created Polycefin - nanoconjugate of natural polymer poly-beta-malic acid (poly- -malic acid, PMLA) from microorganism Physarum polycephalum, with antisense CHI3L1 mRNA oligonucleotides or antibodies against the CHI3L1 protein, and other functional modules. Polycefin will be used for the treatment of glial tumors for antisense inhibition of protein target. Basic modules, necessary for further synthesis of Polycefin, were obtained as a resut of the fullfilment of first project stage. Procedures of PMLA activation and bonding of functional modules to polymeric matrix were worked out. Recombinant CHI3L1 protein was synthesized in prokaryotic system and purified by affine chromatography on Ni-NTA-agarose. Native CHI3L1 protein was isolated from conditioned medium of human osteosarcoma cell line MG-63 and purified by affine chromatography on heparine-sepharose.
