The research aim was to improve the treatment methods for autoimmune diseases (AIDs) based on the study of genetic apparatus state and its functional manifestation in hematopoietic stem (HSCs) and mesenchymal stem cells (MSCs). In the research there were used modern methods of the studies such as: cytofluorimetric, immune enzyme and PCR-analysis method. There was developed the experimental model of one type of autoimmune diseases, especially an acute type, s graft-versus-host disease (GVHD). There was found the reduced content of hematopoietic stem (HSCs) and mesenchymal stem (MSCs) cells, as well as T-regulatory (T-reg) cells against bone marrow hypoplasia, developing as a result of immune disease as GVHD. Taking into account the possible use for treating GVHD the preparations of fetal origin, in the work there were assessed the phenotype characteristics and state of genetic apparatus of HSCs and MSCs of fetal liver of different gestation terms prior to and after cryopreservation. It has been established the correcting effect of FLCs as for the cells of stem compartment of the recipients with GVHD, as well as Treg cells with suppressive activity and reduced the pathology severity. The highest correcting effect was inherent to cryopreserved FLCs of late gestation terms, which in greater extent stimulated the expression of genes, responsible for hemopoietic activity of HSCs and immune suppressive of MSCs. The studies are or priority character and enable not only the widening of our notion on the mechanisms of development of autoimmune conflicts on cell and molecular levels, but and substantiate the expediency of the directed increase in the efficiency of treatment of one of the forms of autoimmune diseases, GVHD with co-transplantation of histoincompatible bone marrow with FLCs.
