Komisarenko Institute of Endocrinology and Metabolism оf the Academy of Medical Scienses of Ukraine" popup.description2 Object of research: patients' blood and samples of the unmodified and pathologically changed thyroid tissue, removed in patients for different thyroid diseases. Aim: To study the role of lysosomal cysteinic cathepsins in apoptosis of thyroid cells in patients with different thyroid pathology. Methods: biochemical, statistics: spectrophotometer, photoelectrocolorimeter, pH-meter, centrifuges, thermostat, calculator. Results: Identification activity of lysosomal cysteinic cathepsins B, H and L in lysosomal and cytosolic fractions obtained from extranodular unmodified tissue, tissue with signs of pathological changes and tissue of tumors showed that the most essential changes in enzyme activity occur in carcinoma and thyroid oxyphilic cellular adenomas. The degree of an increase in cathepsins activity in lysosomes from the tissues of diffuse toxic goiter depends on the severity of thyroid epithelial hyperplasia, from the tissue of papillary carcinomas – on the category T tumors, their follicular structure, presence of capsule, proliferation of tumor cells in the gland or in the adjacent soft tissue, an invasion to lymphatic vessels. Results concerning the nature of changes in the activity of individual cathepsin cytosol may indicate a decrease in intensity of way of lysosomal apoptosis in diffuse toxic goiter tissue, in tissue with chronic thyroiditis, in adenoma tissue solid structure, to activate it in tissue oxyphylic thyroid adenomas and no significant changes in tissue papillary carcinomas.Theoretical and practical novelty: The data obtained on complex and controversial changes in cathepsins like activity of patient’s blood argue disclaim that the its definition can be a reliable additional criteria for differential characteristics of the diseases. The novelty and advantages over the analogs: difference between this study and existing ones and its novelty is the detailed study of changes in activity of cysteinic cathepsins B, H and L in lysosomes and the cytosol, isolated not only from tissue of cаncer but also from pathologically changed thyroid tissue, and also an analysis of revealed changes depending on a number of clinical, functional and pathomorphological characteristics of the diseases. A similar analysis of the prototypes are absent. The effectiveness of implementation: data are published in scientific periodicals and published as scientific materials of conference. Field of use: scientific and practical institutions of endocrinologic and oncologic profiles, medical universities.
