Addition of CHI3L1 to the cell culture medium stimulated cell division and, conversely, addition of CHI3L2 inhibited cell growth. 293_CHI3L1 cells which stably produce CHI3L1 protein, are able to anchorage-independent growth in soft agar and to form foci. These features evidenced about oncogenic properties of CHI3L1. 293_CHI3L1 cells generate ROS more rapidly when exposed to menadione than that of the control 293 cells, thus a producing CHI3L1 glycoprotein by these cells leads to modification of their redox properties. Simultaneous addition of both proteins to the cell culture medium inhibits the proliferative effect of CHI3L1 protein. Inhibition of cell growth can be attributed to the activation of apoptosis-associated caspases-3 and -7 in 293 cells which were processed solely by CHI3L2, or in combination with CHI3L1. CHI3L2-activated cells unfastened from agar and subjected to shape change to spherical ones like after application of known apoptotic agent staurosporin. The use of specific inhibitors of apoptotic signal-regulated kinase 1 (ASK1) resulted in inhibition of apoptotic influence of CHI3L2 protein on the cells. It can be assumed about ASK1-mediated influence of CHI3L2 protein on the apoptosis development in activated cells. It has been revealed that CHI3L2 initiates RI3K signaling cascade by activating of AKT phosphorylation in 293 cells and U373 cells.
