The methodology for gel-filtration with identification of individual fractions of kappa- and beta-caseins has been developed. The methodology for casein micelles producing in the water-protein-acidic polysaccharide system was proposed. The proteolytic activity, the resistance to phages, antagonistic activity and biochemical properties including the production of lactate, diacetyl, acetoin, CO2, the resistance to sodium chloride and antibiotics, of 45 lactic acid bacteria strains. The localization of proteolytic enzymes in lacococcal cells was established. The criteria for their selection that are based on the extra- and intracellular proteases ratio were justified. The proposed model of employing the proteolytic systems from the selected strains of Lactococcus lactis subsp. lactis, Lactococcus lactis subsp. cremoris, Lactococcus lactis biovar. diacetylactis, milk-clotting enzymes and the combination thereof, resulted in production of alphaS- and beta-casein derived products.
