A high risk of disease was in 30% of patients, interim - in 18.5%, and low - in 51.5% of patients, according to Sokal prognostic scale. Cytogenetic studies (karyotyping of bone marrow cells and fluorescence in situ hybridization - FISH) revealed clonal evolution in 31 cases including 8 patients with cytogenetic changes observed before the start of imatinib treatment, and 23 patients after beginning of treatment with tyrosine kinase inhibitors. Before the use of imatinib additional cytogenetic aberrations have been typical in late phases of CML, while on cytogenetic progression complex karyotype with three or more additional aberrations have been often observed. Analysis of mutation status was held in 29 patients with resistance to imatinib. Point mutations of BCR-ABL gene was found in 10 cases: 3 patients with E255K/V, 2 patients with F359V, one patient with both mutations T315I, M351T, Y253H, G250E and H369R. The combination of clonal evolution and point mutations were found in half of patients, which may indicate a lack of connection between these processes. However, analysis of response to treatment with tyrosine kinase inhibitors of 2nd generation suggests a question worthy of further studies.
