The diagnostic methods comprising different variants of polymerase chain reaction (PCR) analysis, restriction fragment length polymorphism (RFLP) as well as modifications in the methods of real-time PCR in the embodiment of TaqMan were developed for analysis of allelic variants of MTHFR, F5, F2, eNOS, ACE , ESR, ITGA2 and CYP2C9 - stroke susceptibility candidate-genes. The synthesis and development of the design of PCR primers for exon sequences of different genes MTHFR, F5, F2, eNOS, ACE , ESR, ITGB3 and CYP2C9 and fluorescently - labeled probe for real time PCR of gene F5 and F2 in the embodiment of TaqMan were carried out. Developed methods were tested on reference DNA samples with the relevant mutations, providing independent analysis of alternative methods. The panel of DNA samples of individuals with known mutations in genes MTHFR, F5, F2, ACE, eNOS, ESR1, ITGA2 and CYP2C9 was established. On the basis of the developed techniques developed prototypes of test systems for molecular diagnosis of the genetic susceptibility to stroke that do not require expensive equipment and can be implemented in specialized medical institutions.
