The aim of the research project was a development of organocatalytic enantioselective methods for the synthesis of trifluoromethylated acyclic aminoketones, aminonitriles and aminoacids. To solve this problem, asymmetric reactions of acyclic trifluoromethylated ketimines with hydrogen cyanide (Strecker reaction), and ketones (Mannich reaction) in the presence of chiral organic catalysts have been studied. Chiral secondary amines (pyrrolidine derivatives), L-proline and thioureas, functionalized by chiral amine fragments have been synthesized and utilized in asymmetric transformations. The relationship between the structure of the substrate (the presence of electronewithdrawing ester function at nitrogen atom), organocatalyst nature and reactivity and enantioselectivity of asymmetric reactions has been established. Obtained compounds have been used as a "building blocks" for the synthesis of enantiomerically enriched chiral trifluoromethyl-containing aminoacids.
