The purpose - To prove experimentally the new approach to treatment of skin melanoma patients by carrying over tumor-specific immunoreactivity by transferfactor polypeptides (TFP). Methods of research - immunological, flow cytometry, experimental, a method of cell culture, statistical. The equipment: centrifuges ОPN-3, ОPN-8, Т 23; a microscope; flowing cytometer FACScan; the automatic sampleres; termostates; the СО2-incubator. On the developed technology low-molecular samples nonspecific and melanoma-specific ТFP in different experimental systems are received. In vіtro ТFP samples specific to antigens melanoma В16, irrespective of a source of their reception, induce more expressed answer of СВА and С57BL/6 mice lymphocytes on melanoma В16 cells compared with not specific samples of a similar origin. Samples embryonal ТFP, irrespective of their specificity and concentration, reduce apoptotic lymphocyte activity mice C57BL/6, rat ТFP such action do not possess. The specific rat ТFP strengthen lymphocyte cytotoxicity CBA mice against melanoma В16 cells and do not influence on such at mice C57BL/6; embryonal ТFP raise lymphocyte cytotoxicity СВА and С57BL/6 mice, and melanoma-specific ТFP are more effective, than nonspecific. In vіvo after disposable introduction ТFP to mice С57BL/6 melanoma-specific samples essentially strengthen proliferative and cytotoxic lymphocyte activity in comparison with nonspecific samples. Thus, melanoma-specific ТFP, irrespective of an origin source, are capable to transfer іn vіtro and іn vіvo immunoreactivity to melanoma В16 antigens to non-sensitized lymphocytes. The melanoma-specific ТFP sample received from rat's embryonal cells, on model passive dissemination melanoma В16 at mice С57BL/6 at disposable preventive introduction creates protective antimetastatic effect which is shown by frequency decrease of dissemination on 37,5 %, reduction of quantity and volume of pulmonary metastasises, a high index of depression of dissemination (85 %). Preventive introduction embryonal samples ТFP promotes essential reduction of quantity and volume of metastasises, however does not influence frequency melanoma В16 dissemination at mice С57BL/6. The depression index of dissemination at use embryonal specific ТFP - 46 %, ТFP nonspecific - 95 %. Thus, samples rat specific ТFP and embryonal nonspecific ТFP possess the expressed protective action at such mode of use at mice C57BL/6 with melanoma В16. At methronomic mode at mice С57BL/6 with melanoma В16 any of applied ТFP samples does not influence average life expectancy of animals. Results of research testify that the received samples melanoma-specific ТFP are perspective for the further studying of their antineoplastic action at other schemes of application, and, first of all, after removal of a tumor for the purpose of an experimental substantiation of expediency of their application in complex treatment of skin melanoma patients.
