Aim of research: to determine the role of hypoxia inducible factors (HIF) family in mechanisms of cardial and cerebral cytoprotective effects of periodic hypoxic hypoxia (PHH), to define optimal PHH mode in order to obtain simultaneous cytoprotective effect in hypoxic/ischemic damage vulnerable organs. Methods of investigations: physiological, biochemical and molecular. Physiological and biochemical investigations of hypoxical preconditioning were conducted on Wistar rats, part of which was adapted to middle mountains conditions. It was revealed, that long adaptation of rats to middle mountains conditions causes adaptive metabolic changes that are manifested with: decrease of aerobic metabolism, intensifying of carbohydrate exchange with development of hypoglycemia, increase of carbohydrate and fatty NAD-dependent substrates of respiratory chain utilization by mitochondries with increased indexes of ADP-stimulated respiration. Also such adaptation induces increasing of carbohydrate and fatty substrates oxidation in liver under normal and acute hypoxia conditions. Character and severity of acute hypoxia induced metabolic reaction has changed as a result of adaptive metabolic changes in rats that are long time living in middle mountains conditions. The developing stress reaction was hypoglemic rather than hyperglemic and following reduction of aerobic metabolism was limited. Transcriptional activation HIF-1? gene was discovered in both heart ventriculars during hypoxical preconditioning. This can indicate its important value as fenotypical cell change assisting factor. This mechanism wasn't activated in right ventricle of mature rats heart and also during hypoxia adaption, thus hypoxical reactivity of miocardium was decreasing. It was discovered prevalence of HIF-3? gene transcriptional activation in left ventricle of miocardium during hypoxical preconditioning.and also marked increase of that activation in mature rats. This can be described as defence mechanism designed to limit HIF-1-dependent induction of target genes. During hypoxial preconditioning in mature animals HIF-3? /HIF-1? ratio in left ventricular was considerably decreased, promoting their fenotypical change. Chronic hypoxia may limit greatly this ability through considerable stimulation of HIF-3? subunit expression, that is much less susceptible to age-related changes.
