Research goal: An experimental investigation of antitumor effects and cellular and molecular mode of action of the recombinant polypeptide EMAP-II, gold nanoparticles and their nanobiocopmositions on the prostate cancer. Research methods: biochemical, biological, morphological. Results: The recombinant polypeptide EMAP-II, 15 nm gold nanoparticles and their nanobiocopmositions injected subcutaneously significantly inhibited growth of the human prostate cancer xenografts in CBA mice nephrosubcapsular model that has been confirmed by histopathology examination. Destructive changes of malignant epithelium induced by combination of EMAP-II, gold nanoparticles or their nanocomposition were more pronounced than those of separate administration of both preparations. Nanocomposition of EMAP-II with Dextran-70 in concentration of 10-3 mcg/ml inhibited significantly growth of cultured LnCaP cells and induced apoptosis that was revealed using TUNEL technique, while monocomponent EMAP-II preparation did not caused such an effect as well as 3Н –thymidine incorporation into the cells. 3Н–thymidine incorporation into cultured human prostate cancer explants exposed in vitro to the gold nanoparticles preparation (1 mcg/ml) was significantly decreased, and apoptosis was enhanced. It was concluded that recombinant polypeptide EMAP-II, 15 nm gold nanoparticles and their combination at ultra-low dosage are able to inhibit growth of human prostate cancer xenografts in mouse model, to make an impact on proliferation and apoptosis in cancerous tissue that should be taken into account in developing the new therapeutic modalities in prostate cancer. Priority: It was found antitumor effect of gold nanoparticles (15 nm) and their combination with ЕМАР-II cytokine with regard to androgen-dependent human prostate cancer xenografts and decrease of DNA synthesis as well as enhancement of apoptosis in cultured tumor tissue exposed to gold nanoparticles. It was shown destructive influence of the nanobiocomposition of EMAP-II with Dextran-70 on cultured cancerous LnCaP cells. Basic importance of the work is determined by further understanding of biological properties of gold nanoparticles, EMAP-II cytokine and nanobiocompositions. Efficacy of implementation: The results will be useful for further research to promote development of a new effective remedies for androgen-dependent prostate cancer. Advantages in comparison to analogues: It is proposed a new method of antitumor impact on androgen-dependent prostate cancer which includes administration of recombinant polypeptide ЕМАР-II or its combination with gold nanoparticles.
