For this analysis to be conducted the design of specific LPO і RPO oligonucleotide zonds and promers was developed considering the temperature of melting of the LHS та RHS sequences, analysis of their secondary structure and probability of the tautomerization between guanine and timidine residues. The composition and the concentration of the key components for the ligase and polimerase chain reaction were optimized - LPO та RPO zonds and primers, thermostable DNA-ligase and thermostable DNA-polimerase. Temperature, time regimes for the ligase and polimerase chain reaction were optimized. The developed methods were verified with the DNA samples of different allel variants of the investigated polymorphisms using referred samples independently analyzed by alternative methods. Those methods can be used for development of test systems of DNA markers for genetic predisposition to cardiovascular diseases.
