Goal - Investigate the genetics and epigenetic mechanisms of neuroblastoma progression and determine factors of the disease prognosis in children. Methods - molecular-genetic, cytogenetics, ulti-color flow cytometry, morphological, biochemical, statistical. Equipment - 7500 Real-Time PCR Systems "Applied Biosystems" (USA), flow cytometer FACSCalibur "Becton Dickinson"(USA), fluorescence microscope Imager M1 "Axio Zeizz" (Germany), centrifuges "ОПН-3", scales WPS. It was established that the risk of neuroblastoma depends on inherited polymorphic variants of the MTHFR gene. We observed a threefold significant increase of neuroblastoma risk in individuals with Т677Т genotype. It was found that MTHFR C677T gene polymorphism has no independent prognostic value for clinical outcome of the disease and response to therapy in patients with neuroblastoma. The obtained results suggest that increased level of MDM2 gene expression, chromosome 1p36 deletion and tumor diploid status are important markers of neuroblastoma prognosis. Defined markers could be included in molecular-genetic component of stratification system of neuroblastoma patients. The obtained results suggest that subsequent research in this area are very promising, especially epigenetic mechanisms of gene regulation. Product Description popup.authors Іонкіна Н.В. Свергун Н.М.
