Objective was to identify the main stages of chemical methods of viral inactivation in the course of technological obtaining blood protein preparations using pre-fractionation methods and affinity chromatography on macroporous silica carriers. Methods: fractionation with virusinactivation chemical agents, biospecific and gel-permeation chromatography, centrifugation, ultradiafiltration, electrophoresis, and others. As an outcome of the work are obtained following scientific-practical results: the first time in processing technology to separate blood plasma protein products using Kohn fractionation, salt and PEG-reprecipitation and chromatographic purification on biospecific dye - siliceous macroporous sorbents, introduced thiocyanate method to inactivate viruses use for viral inactivation in one flowsheet different mode of action of chemical agents (alcohol , PEG - deposition, thiocyanate, solvent-detergent) due to their synergy increases the antiviral effect of the final preparative production of protein drugs. Designed in a simple using photometric express - method for determination of thiocyanate in plasma processing products, allows measuring the concentration range 0,0005-0,005 mol/l and studied experimentally proven otherwise negative impact study inactivating agents on the activity of clotting factors and fibrinolysis. Investigated the effect of virusinactivation reagents on sorbtion properties of macroporous silicon - inorganic sorbents different specificity; unsubstantiated possibility of thiocyanate not only for antiviral treatment, but also for the specific protein components with elution chromatography sorbents for the removal of thiocyanate from protein solutions proposed use of the methods of precipitation, dialysis, ultradiafiltration, gel-permeation chromatography; experiment proved no residual content virusinactivation agents in final protein products.
