In this paper we have been involved immunocytochemistry, cytoflyuorimistry, molecular biological, biochemical and statistical methods of analysis. Objective: to characterise the influence MSKH proliferation, survival, receptor profile and the ability to form multicellular spheroids tumor cells in vitro. Objectives: the determination of kinetic parameters of growth of human tumor cells in culture with co-cultivation with MSKH, assessment of the level of expression of some oncogenic markers tumor cells under the influence of interferon alpha, MSKH and a comparative analysis of the above parameters at different stages of cultivation in monolayer and spheroid culture. The comparison was performed on the expression level of estrogen receptors, epidermal growth factor receptor, cytokeratins and E-cadherin. As a result of studies and comparison of the data obtained during two years of research, it was determined that the T-lymphocytes, in general, adversely affect the growth of transformed cells MCF-7 in vitro. This is manifested in the activation of the expression of ER and EGF-R, stimulate proliferation of cells and reduction of adhesion properties. Thus it can be assumed that the cytokines secreted by T-cells in a medium (IL-2, IL-4, INF-?, TNF) inhibit DNA synthesis cells MCF-7, delaying them in G0/G1, lowering the level of apoptosis in cell population. Stimulated both ER and R-EGF play a role of the transcription factor and may contribute to the transcriptional activity of the cytokine genes, increase the activity of eNOS and affect the activity of p53. Simultaneously to the environment of human bone marrow MSCs have a cytostatic effect on the tumor population holding the number of living cells in suspension or in adhesion control below. By incubation with a medium-MSCs percentage of EGF-R+ cells increased from 66,6% (control) to 83,3%, the number AE1/AE3+ cells up to 100% and keeps the number of Pan+ cells at the level of 100%. According to our results, the percentage of cells expressing E-cadherin, after incubation in medium from MSCs reduced from 50% (control) to 17,4 %. It appeared that the secretory activity of tumor cells in 2D-culture system of 2,39 times larger than the 3-dimensional.
