Object of study: rats and guinea-pigs, their organs, tissues and cells in models of carbonyl stress, such as glycerol stimulated rhabdomyolysis, streptozotocin-induced diabetes and albumin induced asthma. The aim of the work was to clarify the mechanisms of various endogenous aldehydes action, their role in the pathogenesis of diseases with carbonyl and / or oxidative stress, and also to develop the way for prevention and treatment these diseases. It has been conducted the investigation of the effectiveness of a number of drugs which are able to capture formaldehyde and other carbonyl compounds. Aldehydes hyper production involving in the course of pathological processes and the influence of formaldehyde and other aldehyde compounds on the activity of key enzymes of metabolism were investigated too. It has been applied biochemical (chromatographic, centrifuge, spectrometric, etc.) and toxicological methods. Active aldehydes react with thiols compounds (cysteine, glutathione) and amino acid residues of proteins that was the key for the development of a new drug "Correctine 2" containing glycine, lysine and N-acetylcysteine. This drug was used for the normalization of endogenous aldehydes level and for the reducing of post synthetic protein modifications under aldehyde overproduction in the body. In in-vivo rat's experiments on a model of oxidative stress - glycerol stimulated rhabdomyolysis, on a model of hyperglycemia - streptozotocin-induced diabetes it has been recorded the strong carbonyl and oxidative stress. Same processes were detected under chronic albumin-stimulated asthma on guinea-pigs. There were several fold increase in the total content of aldehydes and post synthetic protein modifications, for example N(6)-Carboxymethyllysine. This drug's administration to animals led to the reduction of aldehydes level up to the one in the control group. N(6)-Carboxymethyllysine concentration and other indicators of protein modifications were decreased with "Correctine 2" too. Thus, the drug "Correctine 2" is a perspective formula for its application in complex treatment programs of such chronic diseases that have strong oxidative stress and hyperglycemia.The obtained results reveal new aspects of the metabolism of aldehydes and methods of their overproduction normalization in the body.
